Tier List: Cell lines

Here is the Cell Line Tier List.

S Tier: The Pantheon (Every Biologist Must Know)

These are foundational to modern medicine, biopharma, and cell biology. You cannot pass a molecular biology qualifying exam without knowing them.

  • HeLa: The progenitor of them all. Henrietta Lacks’ immortal cervical carcinoma; ubiquitous, indestructible, and responsible for countless Nobel prizes.
  • HEK 293 / 293T: The viral packaging and protein expression kingpin of biotechnology and synthetic biology.
  • CHO: Chinese Hamster Ovary cells. The billion-dollar biopharma engine that churns out the majority of therapeutic monoclonal antibodies.
  • Jurkat: The undisputed sovereign of T-cell biology. Isolated from a teenage leukemia patient in the 1970s, almost everything we know about T-cell receptor (TCR) signaling, T-cell activation, and early apoptosis biochemistry was mapped out in Jurkat suspension cultures.
  • MCF-7: The queen of oncology. Derived from the pleural effusion of Sister Catherine Frances Mallon in 1970. It is the gold standard ER+/PR+ luminal breast cancer line. If a paper discusses estrogen signaling, endocrine therapy resistance, or SERMs (like tamoxifen), MCF-7 is guaranteed to be in Figure 1.

A Tier: The Heavyweights (Found in Nearly Every Cancer Paper)

Ubiquitous across oncology, high-throughput drug screens, and mechanistic studies. Most life scientists encounter these regularly.

  • MDA-MB-231: The undisputed king of Triple-Negative Breast Cancer (TNBC) models. Highly invasive, mesenchymal, spindle-shaped, and the default choice for metastasis and transwell invasion assays.
  • A-549: The absolute gold standard for Non-Small Cell Lung Cancer (NSCLC, KRAS-mutant) and respiratory infection models (influenza, adenoviruses, SARS-CoV-2). Grows rapidly and reliably.
  • LNCaP & PC-3: The classic prostate cancer duo.
    • LNCaP: Androgen-dependent, PSA-secreting, AR-positive workhorse.
    • PC-3: The high-grade, AR-negative, bone-metastatic, castration-resistant bruiser.
  • PANC-1: The most cited pancreatic ductal adenocarcinoma (PDAC) cell line. Harboring classic KRAS and TP53 mutations, it is the primary workhorse for pancreatic cancer chemotherapy resistance.
  • MCF-10A: The standard non-tumorigenic “normal” control line for mammary epithelium. If someone makes an oncogenic claim in breast cancer, reviewers will demand, “Did you show this doesn’t happen in MCF-10A?”
  • U2OS / HepG2 / K562(External benchmarks): The universal standards for cell biology microscopy (U2OS), hepatotoxicity/liver screens (HepG2), and CML/leukemia suspension studies (K562).

B Tier: Specialized Champions (Essential Subtype Models)

Standard-bearers within their specific niches. In their respective subfields, they are essential.

  • SK-BR-3: The premier HER2-amplified breast cancer line. Trastuzumab (Herceptin) and ADC assays rely on this as their primary model.
  • A-375: The poster child for malignant melanoma driven by the famous BRAF V600E mutation. Essential for studying MAPK/ERK signaling and RAF/MEK inhibitors.
  • 786-O: The default clear cell renal cell carcinoma (ccRCC) model. It carries a homozygous VHL mutation, making it the classic line for studying HIF-1α/2α deregulation and pseudo-hypoxia.
  • SK-OV-3: The most widely published ovarian adenocarcinoma cell line. (Though debated whether it truly reflects high-grade serous histology, it remains an indispensable screening tool).
  • DU145: The “middle child” of prostate cancer. Moderately metastatic, androgen-insensitive, brain metastasis-derived. Rounds out the tri-panel alongside PC-3 and LNCaP.
  • MDA-MB-468: Essential basal-like TNBC line. Famous for carrying heavy EGFR amplification and a PTEN deletion, contrasting with MDA-MB-231.

C Tier: The Panel Fillers & Second-String Options

Respected, well-characterized, but typically drafted only when Reviewer 3 asks for “a second or third cell line to confirm findings.”

  • BxPC-3: High utility in pancreatic cancer because it is KRAS wild-type—an anomaly in PDAC used specifically as a negative control against KRAS-mutant lines like PANC-1.
  • HCC1937: Crucial for germline BRCA1-mutant breast cancer research and PARP-inhibitor resistance studies, but finicky to culture and strictly niche.
  • MG-63: A reliable osteosarcoma line, but lives perpetually in the shadow of U2OS and Saos-2.
  • BT-549: A secondary mesenchymal TNBC cell line. Almost never the headline model; primarily used to back up findings from MDA-MB-231.
  • A-172: A viable glioblastoma line, but largely overshadowed by the dominant U-87 / U-251 glioblastoma lineage.

D Tier: The Niche / Redundant Benchwarmers

Lines that are rarely picked as primary models today due to changing technologies or better alternatives.

  • T98G: A glioblastoma line famous historically for resting in a reversible G1 arrest state under low serum. It has high MGMT expression (conferring temozolomide resistance), but outside of niche cell cycle or MGMT DNA-repair papers, modern neuro-oncology labs rarely default to it.

F Tier: The Disgraced, Cross-Contaminated, and Obsolete

The cautionary tales of cell culture history. Papers relying on these without addressing their flaws are immediate red flags.

  • MDA-MB-435: The Great Impostor. Used for nearly three decades as a flagship metastatic breast cancer line in hundreds of funded studies. Microarrays and STR profiling eventually proved it was an M14 melanoma cross-contamination.
  • HEp-2 & KB: Long published as “laryngeal carcinoma” and “oral epidermoid carcinoma”—both were exposed decades ago as aggressive HeLa overgrowth.
  • Chang Liver: Originally marketed as “normal human liver cells”—turned out to be HeLa yet again.
  • ATCC U-87 MG (The Identity Crisis): A cautionary tale in neuro-oncology. In 2016, DNA profiling proved the widely distributed ATCC stock of U-87 MG did not match the original glioblastoma patient tumor established at Uppsala in 1968 due to a historical cross-contamination event (though labs still use it out of sheer inertia).
